Long R3 IGF-I
An 83-amino-acid IGF-1 analogue engineered with an Arg3 substitution and a 13-residue N-terminal extension to greatly reduce IGF binding-protein affinity.
What the literature describes
Long R3 IGF-I was originally engineered for biopharmaceutical cell culture, where it is a more potent alternative to insulin in serum-free media. The Arg³ substitution and N-terminal extension sharply reduce binding to IGF binding proteins, extending free-fraction half-life relative to native IGF-1. The underlying IGF-1 literature reports hypertrophy driven by a combination of satellite-cell activation and increased protein synthesis in differentiated myofibres, with proliferative effects mediated through PI3K/Akt signalling and downregulation of p27^Kip1.
